Casein is a complete protein that contains all of the essential amino acids required by our bodies to function. In its purest form, casein is a white-colored solid with no taste. All mammals produce casein as a component in milk for their offspring. As a protein supplement, casein supplies our muscles with the full range of amino acids required for exercise recovery and strength building.

After a challenging workout, our bodies repair the small tears created in our muscle fibers to build them back to become bigger and stronger.

Proper sleep and adequate protein intake are critical for muscle recovery. Casein products work by providing an extra boost of protein to support muscle tissue. In its isolated form, casein is used as a primary component in cheese-making and as a protein supplement. Bodybuilders may ingest casein products immediately after a workout or before going to bed to promote exercise recovery. Casein is also used to create infant formulas as a substitute for breast milk.

In nature, casein exists as a molecule that's suspended in a surrounding liquid. This structure is called a micelle. You may picture a micelle as an intact little bubble, mixed into a solution. There are four subtypes of casein. The first three casein subtypes are calcium-sensitive all except for k-casein.

Once k-casein is metabolized, the micelle curdles into an insoluble mass. This initial step of digestion actually transforms casein into a form that's more resistant to breakdown.

Because micelles require several steps for disassembly, casein is considered a slow-digesting protein. For adults, a casein-based protein shake combined with regular resistance training may contribute to bone health and muscle building. A cup of milk has about milligrams mg of calcium, much of which is found within the casein micelles.

Most adults require between 1, mg to 1, mg of calcium per day. In addition to serving as a complete protein, casein is used by bodybuilders because of its slow digestibility. Soy protein and whey protein are released into the bloodstream quickly, while casein takes about six to seven hours to digest fully.

Casein's reduced rate of digestion steadily supplies amino acids to damaged muscle tissue for an extended time after a workout, promoting better recovery. Slow digestion also contributes to satiety, helping you feel full for longer and curb unhealthy food cravings. Cow's milk is one of the most common food allergens , which can pose a problem for formula-fed infants.

Milk allergies usually start during infancy or early childhood, but they can also develop later in life. If a baby is allergic to cow's milk, their healthcare provider may suggest a hydrolyzed casein-based formula. Although its bitter taste isn't always preferred, hydrolyzed casein can help babies with allergies get the nutrients needed during critical growth periods. If you have an allergy to milk, ask your healthcare provider about whether you should have testing to detect the specific proteins responsible for your allergy.

It's possible to be allergic to other proteins in milk, like whey, but not casein. The individual families of casein proteins were identified by alkaline urea gel electrophoresis. Each of the four different caseins may have a variety of numbers of phosphate groups attached through their serine or threonine residues. Another source of variability in caseins is genetic polymorphism. Caseins are structurally classified as natively or intrinsically disordered proteins which is different from random coil conformation found in some globular proteins [ 76 , 77 ].

Caseins proteins possess very little three dimensional structure but possess some secondary structure [ 79 ]. Casein proteins are homologous in all the species as has been found by various protein and gene sequencing studies [ 80 ]. The proportion of various caseins varies widely.

All species form colloidal casein micelles for the transport of calcium and phosphate. Casein micelles of most species appear quite similar at the ultra structural level. Casein proteins are important nutritionally because of their high phosphate content due to which they bind significant quantities of calcium and also are rich in lysine which is an essential amino acid in humans.

The properties of milk and various milk products mainly depend on proteins present in it and to some extent on other components like salts, fat, and lactose. Caseins which possess an extraordinary high heat stability make the milk and other milk products highly stable even at higher temperature [ 61 ].

Mammalian milk contains casein micelles that help to provide adequate nutrients to the neonate and at the same time prevent any risk of pathological calcification or amyloidosis. The structural disorderness and the chaperonic property would have been evolutionarily selected to make these molecules ideally suitable to thrive under various environmental insults since the milk is secretory product.

It might be possible that that in addition to casein, many other milk proteins may also have chaperonic function.
